Retinol — mechanism of action and scientific evidence
Retinol is a form of vitamin A that is biologically active in the skin. After application, it undergoes enzymatic conversion: retinol → retinal → retinoic acid (tretinoin). It is retinoic acid — by binding to the nuclear RAR and RXR receptors — that modulates the expression of genes responsible for collagen synthesis, keratinocyte differentiation and the inhibition of metalloproteinases (MMPs), the enzymes that degrade the extracellular matrix.
The effects have been confirmed in numerous randomized clinical trials: regular use of retinol (0.1–1%) over 12–24 weeks leads to a statistically significant reduction in wrinkles, improved texture and increased skin density. Over-the-counter (OTC) products contain retinol; the more potent tretinoin (0.025–0.1%) requires a prescription.
The downside of retinol is its potential to cause irritation — particularly in the first few weeks of use, redness, flaking and a burning sensation may appear. It needs to be introduced gradually and paired with sun protection.
Peptides — mechanism of action and scientific evidence
Cosmetic peptides are short chains of amino acids (usually 2–10). Depending on their structure, they act as:
- Signal peptides (e.g. Palmitoyl Tripeptide-1, Palmitoyl Pentapeptide-4 — known as Matrixyl) — they mimic fragments of collagen, stimulating fibroblasts to produce it.
- Neurotransmitter peptides (e.g. Acetyl Hexapeptide-3/8 — Argireline) — they reduce the tension of facial muscles, working in a way similar to botulinum toxin, but with far lower and reversible potency.
- Carrier peptides (e.g. GHK-Cu — copper tripeptide-1) — they transport the minerals essential for skin repair.
Studies on Matrixyl show increased collagen density and a statistically significant reduction in wrinkles after 12 weeks — although the effects are usually more subtle than with retinol. Peptides, on the other hand, are much better tolerated: they do not cause irritation and can be used by people with sensitive skin, as well as pregnant and breastfeeding women (for whom retinol is contraindicated).
